ABSTRACT

The systematic assessment of cognitive performance of older people without cognitive complaints is controversial and unfeasible. Identifying individuals at higher risk of cognitive impairment could optimize resource allocation. We aimed to develop and test machine learning models to predict cognitive impairment using variables obtainable in primary care settings. In this cross-sectional study, we included 8,291 participants of the baseline assessment of the ELSA-Brasil study, who were aged between 50 and 74 years and were free of dementia. Cognitive performance was assessed with a neuropsychological battery and cognitive impairment was defined as global cognitive z-score below 2 standard deviations. Variables used as input to the prediction models included demographics, social determinants, clinical conditions, family history, lifestyle, and laboratory tests. We developed machine learning models using logistic regression, neural networks, and gradient boosted trees. Participants' mean age was 58.3±6.2 years, 55% were female. Cognitive impairment was present in 328 individuals (4%). Machine learning algorithms presented fair to good discrimination (areas under the ROC curve between 0.801 and 0.873). Extreme Gradient Boosting presented the highest discrimination, high specificity (97%), and negative predictive value (97%). Seventy-six percent of the individuals with cognitive impairment were included among the highest ranked individuals by this algorithm. In conclusion, we developed and tested a machine learning model to predict cognitive impairment based on primary care data that presented good discrimination and high specificity. These characteristics could support the detection of patients who would not benefit from cognitive assessment, facilitating the allocation of human and economic resources.

ABSTRACT

The objective was to evaluate the intake and digestibility of nutrients, ingestive behavior, and performance of goats fed with spineless cactus genotypes resistant to carmine cochineal (Miúda or Orelha de Elefante Mexicana (OEM). Thirty castrated male goats, without defined breed, aged 12 to 14 months, with an average body weight of 19.0±2.8kg, were distributed in a completely randomized design among three treatments (Control - Tifton hay, Miúda, and OEM) and ten replicates; the initial weight was considered as the covariate. The intake of organic matter (OM), neutral detergent fiber (NDF) was highest in the control treatment, while the intake of NFC was higher in the OEM diet. Treatments containing forage cactus showed the highest digestibility of DM, OM, and NFC. The animals fed the control diet spent more time on rumination and total chewing, but the time spent feeding or feeding efficiency, and performance did not differ. The use of spineless cactus genotypes resistant to carmine cochineal (Miúda and Orelha de Elefante Mexicana) in a diet for goats, in the amount of 450g/kg of DM does not interfere with the performance of the animals and improves the digestibility of OM and NFC.(AU)


Objetivou-se com o presente estudo avaliar o consumo e a digestibilidade de nutrientes, o comportamento ingestivo e o desempenho de caprinos alimentados com genótipos de palma forrageira resistentes à cochonilha-carmim (miúda ou orelha-de-elefante-mexicana (OEM). Trinta cabritos machos, castrados, sem raça definida, com idades entre 12 e 14 meses e peso corporal médio de 19,0±2,8kg, foram distribuídos em delineamento inteiramente ao acaso, em três tratamentos (controle - feno de tifton; miúda e OEM) e 10 repetições; o peso inicial foi considerado a covariável. O consumo de matéria orgânica (MO) e de fibra em detergente neutro (FDN) foi maior no tratamento controle; enquanto a ingestão de CNF foi maior na dieta OEM. Tratamentos contendo palma forrageira apresentaram as maiores digestibilidades de MS, MO e CNF. Os animais alimentados com a dieta controle gastaram mais tempo em ruminação e em mastigação total, mas o tempo gasto com alimentação ou a eficiência alimentar e o desempenho não diferiram. A utilização de genótipos de palma forrageira resistentes à cochonilha-carmim (miúda e orelha-de-elefante-mexicana) na dieta de caprinos, na quantidade de 450g/kg de MS, não interfere no desempenho dos animais e melhora a digestibilidade de MO e CNF.(AU)

ABSTRACT

Cathepsin Z (CTSZ) is a cysteine protease responsible for the adhesion and migration of both immune and tumor cells. Due to its dual role, we hypothesized that the site of CTSZ expression could be determinant of the pro- or anti-tumorigenic effects of this enzyme. To test this hypothesis, we analyzed CTSZ expression data in healthy and tumor tissues by bioinformatics and evaluated the expression levels of CTSZ mRNA in the blood cells of prostate cancer (PCa) patients by qRT-PCR compared with healthy subjects, evaluating its diagnostic and prognostic implications for this type of cancer. Immune cells present in the blood of healthy patients overexpress CTSZ. In PCa, we found decreased CTSZ mRNA levels in blood cells, 75% lower than in healthy subjects, that diminished even more during biochemical relapse. CTSZ mRNA in the blood cells had an area under the curve for PCa diagnosis of 0.832, with a 93.3% specificity, and a positive likelihood ratio of 9.4. The site of CTSZ mRNA expression is fundamental to determine its final role as a protective determinant in PCa, such as CTSZ mRNA in the blood cells, or a malignant determinant, such as found for CTSZ expressed in high levels by different types of primary and metastatic tumors. Low CTSZ mRNA expression in the total blood is a possible PCa marker complementary to prostate-specific antigen (PSA) for biopsy decisions, with the potential to eliminate unnecessary biopsies.

ABSTRACT

The objective of this study was to evaluate the metabolic response of native lambs fed Tifton 85 hay (Cynodon dactylon) or Maniçoba hay (Manihot pseudoglaziovii) associated with spineless cactus (Nopalea cochenillifera Salm Dyck). Sixteen Morada Nova lambs were used, with a mean initial weight of 19.36±1.48kg, distributed in a randomized block design, with two treatments and eight replications. Dry matter intake was higher for lambs fed Maniçoba hay compared to Tifton hay, while the body weight at slaughter was not influenced by diets. Regarding the blood metabolites, only the serum concentration of urea was influenced, with higher value observed in the lambs that fed the diet containing Tifton hay. When analyzing blood biomarkers, in the different biweekly collections, a linear increase in the blood concentration of urea, glucose, fructosamine, alkaline phosphatase and sodium was observed, as well as a quadratic effect for serum potassium. The other blood metabolites were not influenced in function of the collection days. The replacement of Tifton 85 hay for Maniçoba hay does not cause negative changes in the dynamics of different biomarkers and presents itself as a promising forage resource.(AU)


O objetivo deste trabalho foi avaliar a resposta metabólica de cordeiros nativos alimentados com feno de capim-tifton 85 (Cynodon dactylon) ou feno de maniçoba (Manihot pseudoglaziovii) associados à palma forrageira (Nopalea cochenillifera Salm Dyck). Foram utilizados 16 cordeiros Morada Nova, com peso inicial médio de 19,36±1,48kg, distribuídos em delineamento de blocos ao acaso, com dois tratamentos e oito repetições. O consumo de matéria seca foi maior nos cordeiros alimentados com feno de maniçoba em comparação ao feno de tifton, enquanto o peso corporal ao abate não foi influenciado pelas dietas. Em relação aos metabólitos sanguíneos, apenas a concentração sérica de ureia foi influenciada, com maior valor observado nos cordeiros que ingeriram a dieta contendo feno de capim-tifton. Quando foi realizada a análise dos biomarcadores sanguíneos nas diferentes coletas quinzenais, foi observado aumento linear nas concentrações sanguíneas de ureia, glicose, frutosamina, fosfatase alcalina e sódio, e efeito quadrático para o potássio sérico. Os demais metabólitos sanguíneos não foram influenciados em função dos dias de coleta. A substituição do feno de tifton 85 por feno de maniçoba influencia positivamente o metabolismo de cordeiros sem causar alterações na dinâmica dos diferentes biomarcadores e se apresenta como um promissor recurso forrageiro.(AU)

ABSTRACT

Chemerin is an adipokine that has been associated with components of metabolic syndrome. It has been described to affect adipocyte metabolism and inflammatory responses in adipose tissue, as well as the systemic metabolism of lipids and glucose. Few epidemiological studies have evaluated classical and genetics cardiovascular risk factors (CVRFs) in the mixed adult rural population in Brazil. Therefore, the present study explored possible associations between CVRFs and chemerin. This cross-sectional study included 508 adults from the rural localities of Lavras Novas, Chapada, and Santo Antônio do Salto in Ouro Preto, Minas Gerais, Southeast Brazil. Demographic, behavioral, clinical, biochemical, anthropometric variables, and 12 single nucleotide polymorphisms (SNPs) linked with metabolic syndrome phenotypes were evaluated for associations with chemerin level. There was a significant association of high triglyceride levels [odds ratio (OR)=1.91, 95%CI: 1.23−2.98], insulin resistance (OR=1.82, 95%CI: 1.03−3.22), age (OR=1.64, 95%CI: 1.08−2.49), and sex (OR=1.99, 95%CI: 1.35−2.95) with high levels of chemerin. High chemerin levels were significantly associated with the genetic polymorphisms rs693 in the APOB gene (OR=1.50, 95%CI: 1.03−2.19) and rs1799983 in the NOS3 gene (OR=1.46, 95%CI: 1.01−2.12) for the AA and GT+TT genotypes, respectively. In the concomitant presence of genotypes AA of rs693 and GT+TT of rs1799983, the chance of presenting high levels of chemerin showed a 2.21-fold increase (95%CI: 1.25−3.88) compared to the reference genotype. The development of classical CVRFs in this population may be influenced by chemerin and by two risk genotypes characteristic of variants in well-studied genes for hypertension and dyslipidemia.

ABSTRACT

The aim of this work was to evaluate the effect of the Rolipram during the maturation of bovine oocytes and gene expression of embryos produced in vitro. Bovine ovaries were collected in slaughterhouse. The COCs were selected and divided into 5 groups: Control 0 time; Control: IVM for 24 hours; Rolipram treatments with IVM blocking for 24 hours in maturation medium containing (100, 150 and 200µM). After 24 hours all groups were reseated in IVM for another 24 hours. Subsequently COCs were subjected to the same IVM system and fertilized, being checked for cleavage post fertilization and for blastocyst. In addition, performed expression of the following genes: Mater, BMP15 and Bax. No difference was found in gene expression. Of oocytes evaluated shortly after follicular aspiration, 79.00% were in GV, GVBD, MI, while 13.40%, were in MII and 7.60%, D/NI. Significant difference was observed in different concentrations (T100, T200 and T150µM) in oocytes that have reached the MII phase compared to control treatments (P= 0.003). Differences were observed in cleavage rate (P< 0.05) between T150 and T200 when compared to the C/24 Group. A high difference was observed on blastocyst rate (P< 0.001) among treatments compared to the control group.(AU)


O objetivo deste trabalho foi avaliar o efeito do rolipram durante a maturação de oócitos bovinos, expressão gênica e embriões produzidos in vitro. Os ovários bovinos foram coletados no matadouro. Os COCs foram selecionados e divididos em cinco grupos: controle 0 tempo; controle: MIV por 24 horas; tratamentos rolipram com bloqueio MIV por 24 horas em meio de maturação contendo 100, 150 e 200µM. Após 24 horas, todos os grupos foram recolocados em MIV por mais 24 horas. Subsequentemente COCs foram submetidos ao mesmo sistema MIV e fertilizados, sendo avaliada a taxa de clivagem e de blastocisto, além da expressão dos seguintes genes: Mater, BMP15 e Bax. Nenhuma diferença foi observada na expressão gênica. Dos oócitos avaliados logo após a aspiração folicular, 79,0% estavam em GV, GVBD, MI, enquanto 13,40% estavam em MII, e 7,60% em D/NI. A diferença significativa foi observada em diferentes concentrações (T100, T200 e T150µM) em oócitos que atingiram a fase MII em comparação aos tratamentos de controle (P=0,3). Diferenças foram observadas nas taxas de clivagem (P<0,5) entre T150 e T200 quando comparadas com as taxas do grupo C/24. Uma grande diferença foi observada na taxa de blastocisto (P<0,1) entre os tratamentos em relação ao grupo controle.(AU)

ABSTRACT

O objetivo deste estudo foi avaliar o efeito da suplementação do diluidor de congelação de sêmen ovino com o flavonoide miricetina contra os danos ocasionados aos espermatozoides. Oito pools de sêmen, obtidos de quatro reprodutores ovinos, foram congelados com diferentes concentrações de miricetina (0, 1, 10, 100 e 1000nM). Após o descongelamento, o sêmen foi avaliado quanto à cinética espermática, à integridade das membranas plasmática e acrossomal, ao potencial de membrana mitocondrial, aos níveis de ROS intracelular, à peroxidação lipídica e à estabilidade de membrana. Amostras tratadas com miricetina 10nM apresentaram menor percentual de células rápidas (P≤0,05), quando comparadas ao grupo miricetina 1000nM. Amostras do grupo controle apresentaram maior (P≤0,05) VAP que o grupo 10nM de miricetina, enquanto amostras criopreservadas com miricetina (10, 100 e 1000nM) evidenciaram maior (P<0,05) BCF, quando comparadas ao grupo controle. O grupo tratado com miricetina 1000nM apresentou maior percentual (P<0,05) de células com peroxidação lipídica, quando comparado ao grupo controle. Em conclusão, a suplementação do diluidor de criopreservação de sêmen ovino com 10 e 100nM de miricetina afeta a cinética espermática sem provocar alterações na estrutura geral do gameta, enquanto 1000nM de miricetina provoca mudanças na cinética associadas à danos peroxidativos.(AU)


The aim of this study was to evaluate the effect of the supplementation of ram semen frozen with extender with the flavonoid myricetin against damage to sperm. Eight pools of semen obtained from four ram breeders, were frozen with different concentrations of myicetin (0, 1, 10, 100 and 1000nM). After thawing, the semen was evaluated for spermatic kinetics, plasma and acrosome membrane integrity, mitochondrial membrane potential, intracellular ROS levels, lipid peroxidation, and membrane stability. Samples treated with 10nM myricetin preserved a lower percentage of rapid cells (P≤0.05) when compared to the 1000nM myricetin group. Samples from the control group presented higher (P≤0.05) VAP than 10nM group of myricetin, while cryopreserved samples with myicetin (10, 100 and 1000nM) showed greater (P<0.05) BCF, when compared to control group. The group treated with 1000nM myricetin had a higher percentage (P<0.05) of cells with lipid peroxidation, when compared to the control group. In conclusion, supplementation of ram semen cryopreservation extender with 10 and 100nM myricetin affects sperm kinetics, without causing changes in the overall structure of the gamete, while 1000nM myricetin causes changes in the kinetics associated with peroxidative damage.(AU)

ABSTRACT

Informações sobre a vascularização da parede folicular e do corpo lúteo equino, associadas à superovulação, são escassas. Com o objetivo de avaliar o efeito superovulatório do extrato de pituitária equina (EPE) no fluxo sanguíneo folicular e luteal, foram utilizadas seis éguas Puro Sangue Árabe, em dois ciclos estrais (controle e tratamento). As éguas foram monitoradas diariamente por ultrassonografia modo B, até que os folículos atingissem diâmetro de 23mm (desvio). No ciclo tratamento, as éguas receberam 8mg de EPE, uma vez ao dia, por via IM, até que dois ou mais folículos atingissem o diâmetro entre 32 e 35mm. A ovulação foi induzida com acetato de deslorelina, quando os folículos atingiram, no mínimo, 35mm. No momento do desvio folicular, da indução da ovulação e do último exame pré-ovulatório, foi utilizada a ultrassonografia modo B para medir o diâmetro dos folículos e, no oitavo dia pós-ovulação, para a área do corpo lúteo (CL). Utilizou-se também ultrassonografia com Doppler colorido para avaliar a perfusão sanguínea da parede folicular e do parênquima luteal. No ciclo controle, foi realizado o mesmo procedimento, exceto pelo uso do EPE. Os dados foram submetidos à análise de variância, com nível de significância de 5%. Não foi observado efeito do EPE sobre o número de ovulações, o diâmetro dos folículos, a vascularização da parede folicular e a concentração sérica de estrógeno. Os animais, tratados ou não, apresentaram CLs funcionais, não havendo diferença na área do parênquima ou da vascularização luteal, nem na concentração sérica de progesterona, no oitavo dia após a ovulação. Foi observado que o EPE proporcionou um maior número de folículos subordinados no momento da indução da ovulação do folículo dominante (P ≤ 0,05). Embora esses folículos não tenham chegado a ovular, concluiu-se que o EPE atuou no crescimento de folículos, que podem ser utilizados em outras biotécnicas, como a transferência de oócitos, com maior aproveitamento da reserva folicular de ovários equinos.(AU)


Knowledge about follicle and corpus luteum vascularization associated with superovulation in mares is scarce. Aiming to evaluate the effect of equine pituitary extract (EPE) on superovulation, the experiment was conducted using six mares Purebred Arabian in two estrous cycles (control and treatment). The mares were synchronized, and monitored daily by ultrasound B mode until the follicles reached diameter ≤ 23 mm (deviation). In the treatment cycle, from the deviation, mares received 8 mg of EPE, once a day, intramuscularly, until two or more follicles reached a diameter between 32 and 35 mm. Ovulation was induced with deslorelin acetate when follicles reached at least 35 mm. At the time of follicular deviation, induction of ovulation and final preovulatory exam, it was used B-mode ultrasound to measure the diameter of follicles and on the eighth day after ovulation to measure the area of the corpus luteum (CL); color Doppler was also used to assess blood perfusion of the follicle wall and luteal parenchyma. In the control cycle was performed the same procedure except for the use of EPE. Data were subjected to analysis of variance, with 5% significance level. There was no effect of EPE on ovulation number, diameter of follicles, vascularity of the follicular wall and serum estrogen concentration. The animals treated or not, showed functional CLs, with no difference in parenchymal area or luteal vascularization, or in serum progesterone concentration on the eighth day after ovulation. It was observed that the EPE provided a greater number of subordinate follicles at the time of induction of ovulation of the dominant follicle. Although these follicles have failed to ovulate, it was concluded that EPE influenced the follicles growth, and it can be used in other biotechnologies, with greater utilization of equine ovarian follicular reserve.(AU)

ABSTRACT

Objetivou-se avaliar o efeito do cruzamento entre ovinos nativos no desempenho e características da carcaça. Foram utilizados vinte cordeiros, machos, inteiros, com idade inicial de quatro meses, e peso médio inicial 19,0±5,11, distribuídos em dois grupos genéticos: Rabo Largo (RL) e Santa Inês x Rabo Largo (F1SI x RL), em um delineamento inteiramente ao acaso. Os animais foram terminados em regime de confinamento durante 70 dias. Os cordeiros F1SI x RL apresentaram maiores pesos ao desmame, ao abate, do corpo vazio, das carcaças quente e fria, e maior rendimento biológico. Verificaram maiores valores para os parâmetros de morfometria da carcaça para animais F1SI x RL comparados aos animais Rabo Largo (P<0,05). Os pesos da hemi-carcaça e dos cortes comerciais foram superiores para cordeiros F1SI x RL. Nas medidas obtidas no músculo Longissimus dorsi, os cordeiros Rabo Largo apresentaram valores superiores para espessura de gordura subcutânea. A utilização da raça Santa Inês como base paterna acarreta em melhorias nas características de carcaça de cordeiros Rabo Largo podendo ser indicado em sistemas de produção de carne ovina.(AU)


The aim of study was to evaluate the effect of crossbreeding among native sheep on performance and traits carcass. Twenty intact lambs, male, at four months of age, with an initial weight of 19.0±5.11kg, were distributed in two groups genetics: Rabo Largo (RL) e Santa Inês x Rabo Largo (F1SI x RL), in a completely randomized design. The animals were finished in feedlot by 70 days. The lambs F1SI x RL showed higher weight at weaning, to slaughter, of the empty body, and hot and cold carcass, and increased biological yield. Higher values for carcass morphometry in F1SI x RL animals were found in comparison to Rabo Largo animals (P<0.05). The weight of half-carcass and the commercial cuts were higher for lambs. On the measurements obtained in muscle Longissimus dorsi, Rabo Largo lambs showed higher values for subcutaneous fat thickness. The use of the breed Santa Inês like base paternal may bring in improvements in lambs's carcass characteristics Rabo Largo and may be recommended in sheep meat production systems.(AU)

ABSTRACT

RESUMO A espécie Ocimum gratissimum (Linn.), popularmente conhecida como alfavaca-cravo, é muito utilizada na medicina popular. A planta apresenta inúmeros compostos, sendo o eugenol o constituinte majoritário do seu óleo essencial e o provável responsável pela sua atividade antimicrobiana. O objetivo deste estudo foi determinar o potencial antifúngico e o possível mecanismo de ação do óleo essencial extraído das folhas de O. gratissimum L. (OEOg) sobre cepas-padrão de Candida. Para avaliação da atividade antimicrobiana foi determinada a Concentração Inibitória Mínima (CIM), o efeito do tempo de exposição, o efeito modulador na atividade de antifúngicos (ATF) de uso clínico e a ação do OEOg nas fases de crescimento exponencial e estacionário de leveduras do gênero Candida. O mecanismo de ação do OEOg foi verificado por captação do cristal violeta e avaliação da morfologia microbiana pela técnica de microcultivo. Também foi avaliada a toxicidade do OEOg sobre hemácias humanas. O OEOg apresentou boa atividade antifúngica sobre cepas de Candida, sendo capaz de reduzir a taxa de crescimento das cepas de Candida a partir de quatro horas de exposição, além de ter modulado positivamente a atividade do cetoconazol para C. tropicalis ATCC 13803 e reduzir o número de células viáveis em todas as fases de crescimento microbiano. O OEOg foi capaz de promover o aumento discreto da captação do cristal violeta e provocou alterações na micromorfologia das células de Candida spp., sugerindo que seu alvo de ação seja o envoltório celular. Observou-se baixa toxicidade do OEOg sobre hemácias humanas. Os resultados encontrados mostraram que o OEOg possui boa atividade sobre o gênero Candida, com mecanismo de ação mediado possivelmente pela ocorrência de danos no envoltório celular, além de ter sido observada baixa toxicidade, indicando do OEOg é promissor no desenvolvimento e elaboração de um novo fármaco com potencial atividade para o tratamento de doenças fúngicas.


ABSTRACT Ocimum gratissimum (Linn.) is a medicinal plant popularly known as “wild basil” widely used in traditional medicine. The plant has numerous compounds, and eugenol is the major constituent of its essential oil and likely responsible for its antimicrobial activity. The aim of this study was to determine the antifungal activity and the potential mechanism of action of the essential oil extracted from the leaves of O. gratissimum L. (OEOg) against standard strains of Candida. The following experiments were performed: determination of Minimum Inhibitory Concentration (MIC), determination of the effect of exposure time to OEOg; evaluation of the modulating effect of OEOg in antifungal (ATF) activity for clinical use; determination of the effects of the OEOg on different growth phases of Candida spp; determination of crystal violet (CV) uptake and the microculture of yeast technique. OEOg showed good antifungal activity against Candida, being able to reduce microbial growth during 24 hours of contact and also the number of viable cells at all stages of growth. OE positively modulates the activity of ketoconazole for C. tropicalis ATCC 13803. Increased uptake of CV and also the inhibition of Candidavirulence factors were also observed, which indicates the occurrence of damage in the cell envelope. These findings, coupled with the low toxicity of OEOg on human erythrocytes, indicate that “wild basil” is a promising plant for the development of a new drug with a potential activity to treat fungal diseases.

Avaliou-se o consumo e digestibilidade dos nutrientes em dietas para cabras da raça Saanen arranjadas em um quadrado latino 5x5, alimentadas com cana-de-açúcar em substituição (0, 25, 50, 75 e 100%) ao feno de capim-tifton 85. O experimento foi constituído de cinco períodos de 15 dias (10 dias para adaptação dos animais às dietas experimentais e 5 dias para coleta de dados e amostras). O consumo voluntário foi calculado pela diferença entre o ofertado e as sobras. Utilizou-se o óxido crômico para estimativa da produção de matéria seca fecal. A digestibilidade aparente da matéria seca, matéria orgânica, extrato etéreo e fibra em detergente neutro foi influenciada negativamente com a substituição, enquanto a digestibilidade da proteína e dos carboidratos não fibrosos não sofreu influência. O baixo consumo de matéria seca de rações com cana-de-açúcar torna-se um fator limitante para cabras de média a alta produção de leite.


The intake and digestibility of nutrients in the diets of Saanen goats arranged in a 5x5 Latin square design, fed with sugar cane replacement (0, 25, 50, 75 and 100%) hay Tifton 85 were evaluated. The experiment consisted of 5 periods of 15 days (10 days for animal adaptation to experimental diets and 5 days for data collection). The voluntary intake was calculated as the difference between the offered and leftovers. Digestibility chromic oxide was used to estimate the fecal dry matter production. The digestibility of dry matter, organic matter, ether extract and neutral detergent fiber was negatively influenced, whereas the digestibility of protein and non-fiber carbohydrates were not affected. The low dry matter intake of diets with sugar cane becomes a limiting factor for goats medium for high milk production.

ABSTRACT

RESUMO Piper hispidum é uma espécie pioneira pertencente à família Piperaceae, com importância na medicina popular e na obtenção de óleo essencial. Assim como outras espécies da família, possui poucas informações sobre técnicas de cultivo. O objetivo foi avaliar a propagação via estaquia de Piper hispidum em função do tipo de substrato e estaca em Manaus, Amazonas, Brasil. O experimento foi realizado na Embrapa Amazônia Ocidental. O delineamento utilizado foi inteiramente casualizado em esquema fatorial 3 (estacas) x 5 (substratos), com três repetições de 12 estacas, sendo as estacas (apical, mediana e basal) e os substratos (areia lavada, substrato comercial, solo + esterco de aves, solo + casca de guaraná e fibra de coco). Foram avaliadas: enraizamento (%), número de brotações, comprimento da maior brotação (cm), número de folhas, comprimento da maior raiz (cm), massa seca da raiz (g) e massa seca das brotações (g). Foi realizada análise de variância pelo teste F a 5% de probabilidade e para as médias foi realizado o teste Tukey ao nível de 5% de probabilidade. Recomenda-se utilizar estacas apicais e basais, nesta ordem. Ocorreu interação entre os fatores substrato e tipos de estaca somente para variável número de brotações. Os substratos areia lavada e substrato comercial são indicados para maiores porcentagens de enraizamento com 81,56% e 81,33%, respectivamente. O enraizamento foi superior nas estacas apicais (85,67%) e basais (74,47%). Porém, para esta espécie os substratos solo + esterco de aves e/ou solo + casca de guaraná foram mais indicados quando o objetivo é obter estacas de qualidade.


ABSTRACT Piper hispidum is a pioneer species belonging to the family Piperaceae, with relevance in popular medicine and in obtaining essential oil. As other species of this family, there is little information about cultivation techniques. The aim of this work was to evaluate the propagation through cutting from the Piper hispidum according to the type of substrate and cutting technique in Manaus, Amazonas, Brazil. The trial experiment was conducted at the Embrapa Western Amazon. The design was completely randomized factorial 3 x 5, with three replications of 12 cuttings with types of cuttings (apical, median and basal) and substrates (washed sand, commercial substrate, soil + poultry manure, soil + guarana shell and coconut fiber). After 60 days, the following characteristics were evaluated: rooting (%), number of shoots, length of the largest sprouting (cm), number of leaves, length of the longest root (cm), root dry weight (g) and dry weight of shoots (g). An analysis of variance was performed by the F test at 5% probability and for the averages` comparison the Tukey test was done at 5% level of probability. It is recommended to employ apical and basal cuttings, respectively. There was interaction between the factors and substrate types of cuttings only for the variable number of sprouts. These two substrates, washed sand and commercial substrate, are suggested for higher percentages of rooting with 81,56% and 81,33 %, respectively. The rooting was higher in the apical cuttings (85,67 %) and basal ones (74,47 % ). However, for this species, the substrates soil + poultry manure and/or soil + guaraná shell were most indicated when the goal was to obtain high-quality cuttings.

RESUMO A espécie Arrabidaea chica pertencente à família Bignoniaceae é também conhecida popularmente como crajiru. É Utilizada tradicionalmente no tratamento de enfermidades da pele, inflamações, infecções, dentre outros. O objetivo deste trabalho foi avaliar a produção de biomassa aérea, teor e rendimento de extrato de crajiru em função de diferentes fontes de adubo orgânico em Manaus, AM. As mudas foram formadas através de estacas e mantidas por 60 dias em casa de vegetação. O experimento foi conduzido em esquema fatorial 5x3 com delineamento em blocos casualizados onde os níveis dos fatores foram as diferentes fontes de adubo orgânico: composto (5,0 kg/m2), esterco de aves (3,0 kg/m2), casca de guaraná (4,0 kg/m2), esterco de gado (4,0 kg/m2) e controle (ausência), e os três morfotipos de crajiru. Obedeceu-se o espaçamento de 1,0x1,0 m entre planta e 2,0 m entre blocos. Após 240 dias foram avaliadas a produção de folhas e caules, relação folha/caule, teor de extratos hexânico concentrado em evaporador rotativo obtido pela fórmula: (massa do extrato/massa da amostra total)x100; e o rendimento de extrato estimado pela equação: (%extrato x massa seca total das folhas)/100. Os dados foram submetidos à análise de variância e as médias ao Teste de Tukey (p<0,05). As fontes de adubo orgânico promoveram melhor desenvolvimento das plantas de, exceto para o morfotipo 1. O esterco de aves proporcionou maior produção de biomassa, teor e rendimento de extratos. Dessa forma, sugere-se que a adubação orgânica propicia maior produção de biomassa, teor e rendimento de extratos de A. chica.


ABSTRACT The Arrabidaea chicaspecies belongs to the Bignoniaceae family and is also popularly known as crajiru. It is traditionally used to treat skin diseases, inflammations and infections. The objective of this study was to evaluate the biomass production, the content and the yield of crajiru extract in face of different sources of organic fertilizer in Manaus, AM. The seedlings were formed by cuttings and kept for 60 days in a greenhouse. The trial was conducted in a 5x3 factorial design with randomized block organization where the factor levels were different sources of organic fertilizer: compound (5.0 kg / m 2), chicken manure (3.0 kg / m 2), bark guarana (4.0 kg / m2), cattle manure (4.0 kg / m 2) and control (absence), and the three morphotypes of crajiru. It was obeyed the spacing between the plants (1,0 x1,0m) and the blocks (2,0m) . After 240 days it were evaluated the production of leaves and stems, the leaf / stem ratio, and the content of hexane extracts concentrated by rotary evaporation obtained by the following formula: (mass of extract / mass of total sample) x100; and the yield estimated by the equation extract: (% extract x total dry mass of leaves) / 100. The data were subjected to analysis of variance and means to the Tukey"s test (p <0.05). The sources of organic fertilizer promoted better development of plants, except for the morphotype 1. The poultry manure provided higher biomass, content and yield of extracts. Thus, it is suggested that the organic fertilizer provides greater biomass production, content and yield of A. chica extracts.

ABSTRACT

The objective of this work was to study, through ultrasonographic evaluation, changes in testes and epididymides of clinically healthy, peripubertal and pubertal Santa Inês lambs raised in Brazil. Periodic e valuations of weight, biometric characteristics (scrotal circumference, width and length) and ultrasound examinatio ns of the testes and epididymides of 20 lambs were performed between 84 and 280 days old at intervals of 28 days. Scans were performed in the sagittal, transverse, frontal and oblique planes to evaluate the echotexture of the testicular parenchyma and mediastinum and the tail epididymis as well as the thickness and width of the mediastinum testis. The testicular parenchyma demonstrated a homogeneous echogenicity patter n ranging from low to moderate. The echogenicity of testicular parenchyma increased in direct proportion to animal age, being higher in pubertal lambs when compared to prepubertal at the same age. The mediastinum testis was observed in 100% of the evaluated animals, regardless of the scan plane used, and was classified as diffuse or moderately or highly echogenic. Echogenicity and the thickness of the mediastinum testis increased in direct proportion to animal age. The epididymal tail was presented in hypoechoic relation to the testicular parenchyma. Based on these results, it was concluded that ultrasound is useful tool for selection and morphophysiological evaluation of Santa Inês lambs on peripubertal and pubertal phases, when used in combination with other methods such as semen evaluation.

ABSTRACT

Avaliou-se a integridade de espermatozoides ovinos colhidos e criopreservados em diferentes situações climáticas na região semiárida do estado da Paraíba. As colheitas de sêmen foram realizadas em duas épocas do ano, período seco e período chuvoso, utilizando cinco machos adultos da raça Santa Inês, com histórico favorável de fertilidade. Os ejaculados foram avaliados quanto à motilidade, vigor, concentração e morfologia espermática, após a formação do pool e diluição em Tris-gema, na concentração final de 240x10(6) espermatozoides/mL. Motilidade total e vigor espermático não diferiram entre as épocas seca e chuvosa. Valores de integridade do acrossoma e da membrana plasmática, e o potencial de membrana mitocondrial foram mais baixos (P<0,05) na época com menor índice pluviométrico. Conclui-se que a reprodução de ovinos criados na região do semiárido paraibano sofre ação da condição climática e sugere-se que a criopreservação de espermatozoides ovinos seja realizada no período de maior índice pluviométrico na região.


Ram spermatozoa integrity collected and cryopreserved in different climatic situations in the semiarid region of Paraíba state was evaluated. Semen samples were collected in two periods: a dry and a rainy season, using five mature Santa Inês males, with favorable historical fertility. Ejaculates were evaluated for motility, vigor, concentration and morphology, after a pool formation and dilution in Tris-yolk egg, at 240x10(6)/mL final concentration. Motility and sperm vigor did not differ between dry and rainy seasons. Acrosome integrity, plasma membrane and mitochondrial membrane potential were lower (P>0.05) at the time with less rainfall. We concluded that sheep reproduction raised in semi-arid Paraiba region suffers climatic condition influence and the lowest rainfall period is deleterious to the sperm cells integrity subjected to cryopreservation process. It is suggested that ram spermatozoa cryopreservation be done in period of greatest rainfall in this region.

ABSTRACT

Verificaram-se os efeitos da associação de furosemida e fenilbutazona sobre variáveis hidroeletrolíticas de cavalos antes e após a corrida. Dezenove equinos foram distribuídos em três grupos, de acordo com os protocolos de tratamento. O primeiro grupo, de cinco animais, não recebeu medicação (grupo-controle); o segundo grupo, de sete animais, foi tratado com furosemida, na dose de 1mg/kg, por via intramuscular, até quatro horas antes do páreo; o terceiro, de sete animais, recebeu furosemida, por via intramuscular, e fenilbutazona, por via intravenosa, nas doses de 1,0 e 4,4mg/kg, respectivamente, até quatro horas antes da corrida. Amostras de sangue foram colhidas antes, imediatamente após e duas horas após o páreo, para avaliação da osmolalidade plasmática e das concentrações plasmáticas de sódio, potássio e cloreto. A utilização de furosemida e da associação furosemida e fenilbutazona até 4h antes dos páreos nas dosagens descritas alterou (P<0,05) a osmolalidade plasmática dos equinos, mas não alterou (P>0,05) as concentrações de sódio, potássio e cloreto. Os páreos alteraram de forma fisiológica a osmolalidade plasmática e a concentração sanguínea de K+ devido ao exercício de alta intensidade.


The objective of this study was to verify the effects of furosemide and phenylbutazone association on fluid and electrolyte balance characteristics of horses before and after a race. Nineteen horses were divided into three groups according to treatment protocols. The first group (five animals - control) was not medicated. A second group (seven animals) was treated with furosemide (1mg/kg, intramuscular up to four hours before the race). A third group (seven animals) received furosemide (1mg/kg) and phenylbutazone (4.4mg/kg), both intramuscular, up to four hours before race. Blood samples were collected before, immediately after and two hours after a race to evaluate the plasma osmolality and sodium, potassium and chloride concentrations. The use of furosemide and furosemide plus phenylbutazone up to four hours before the race altered (P<0.05) the plasma osmolality but did not change (P>0.05) the sodium, potassium and chloride concentrations. It was not possible to determine an antagonist effect of phenylbutazone on furosemide, based on fluid and electrolyte balance. Due to the high intensity exercise, the increase in plasma osmolality and potassium concentration was attributed to the race effect.

ABSTRACT

O vírus, contrariamente a outras classes de agentes que afetam o organismo, não são observáveis em microscopia óptica, refletindo-se apenas microscopicamente em efeitos virais (ECPV), que permitem identificá-los por constituírem um conjunto de alterações patognomônicas a nível celular. O aumento nuclear, a variação da morfologia celular, a perda do padrão da cromatina, as inclusões intranucleares ou intracitoplasmáticas ou a multinucleação, são alguns dos ECPV mais comuns. Entre os vírus que afetam a mucosa genital na família Herpesviridae, inserem-se Herpes Simplex 2 e o Citomegalovirus, cujo diagnóstico deverá ter em conta a complexidade dos mecanismos de replicação. Assim, e apesar dos vários métodos para a detecção do vírus, a citologia continua a ser um método viável para o diagnóstico destas infecções virais, sendo que a compreensão do processo de infecção contribui para a correta avaliação microscópica. Este trabalho tem como finalidade efetuar uma revisão bibliográfica sobre o grupo Herpes, focando o Herpes Simplex 2 e o Citomegalovirus, assim como, explicar e relacionar os ECPV, associados ao grupo Herpes , tendo por base os mecanismos de infecção.

ABSTRACT

A closed fracture was performed on the left tibia of 3-month-old Wistar rats weighing 250 to 350 g that were either healthy (N = 24) or made diabetic with alloxan (N = 24) to investigate the effect of alloxan-induced diabetes on the course of bone fracture healing. Histomorphometric analysis of the fracture site was performed at 7, 14, 25, and 35 days. After 7 days, diabetic rats had significantly less cartilage (P = 0.045) and greater fibrous connective (P = 0.006) tissue formation at the fracture site compared to controls. In contrast, marked callus formation was seen in diabetic rats with significant osteogenesis (P = 0.011, P = 0.010, P = 0.010, respectively, for 14, 25, and 35 days) and chondrogenesis (P = 0.028, P = 0.033, P = 0.019) compared to controls. Radiographic analysis revealed a displaced fracture with poor bone fragment alignment and delayed consolidation at these times in the diabetic group. The levels of alkaline phosphatase were significantly higher in diabetic rats at 25 days (P = 0.009). These results suggest that the initial excessive formation of fibrous connective tissue associated with delay in chondrogenesis and osteogenesis may not provide suitable stability of the fractured site, contributing to the inappropriate alignment of fragments and an increase in the volume of callus in later stages of repair. The resulting displaced fracture in diabetic rats requires long periods for remodeling and complete bone consolidation.

ABSTRACT

Incêndios são ocorrências periódicas no Parque Nacional de Ilha Grande, e o fogo é um dos principais fatores de ameaça à sobrevivência da herpetofauna nessa unidade de conservação. Esse trabalho teve como objetivos descrever as reações das serpentes frente ao fogo, identificar as espécies atingidas pelos incêndios e correlacionar a natureza das lesões causadas pelo fogo. As atitudes das serpentes frente ao fogo foram determinadas pela descrição das reações comportamentais apresentadas pelos indivíduos avistados durante os incêndios. A determinação da causa da morte e das lesões foi realizada pela avaliação física e necroscópica dos animais feridos e mortos encontrados em áreas queimadas. As coletas foram realizadas em setembro de 2003, em pontos aleatórios, durante e imediatamente após os incêndios. Foram avistadas frente ao fogo, uma Bothrops moojeni, uma Philodryas patagoniensis e um colubrídeo não-identificado. Uma Micrurus lemniscatus foi encontrada viva apresentando queimaduras profundas e deficiência visual em decorrência das lesões térmicas. Foram coletados cadáveres de quatro B. moojeni, de um colubrídeo não-identificado, e de uma Tamnodynastes hypoconia. As B. moojeni e o colubrídeo não-identificado se encontravam carbonizados, enquanto a T. hypoconia apresentava sinais de afecção respiratória. Vestígios indicaram que os cadáveres das serpentes são uma importante fonte de recurso alimentar nos períodos pós-fogo. Incêndios de elevada intensidade e grandes extensões atingem diretamente serpentes no Parque, inclusive espécies de grande porte como a Bothrops moojeni. A morte de indivíduos pela ação do fogo pode contribuir para o declínio das espécies em longo prazo no Parque Nacional de Ilha Grande.


There are periodic occurrences of in the Ilha Grande National Park (South of Brazil), and it is one of the main menacing factors to the Herpetofauna survival in that conservation unit. The objectives of this study were to describe the snakes’reactions to fire, to identify the species affected by fires, and to correlate the nature of the lesions produced by fire. The reactions of the snakes toward fire were determined by the description of the behavior presented by the individuals observed during wildfires. The determination of mortality causes and correlated lesions were accomplished by physical and necroscopic evaluation of wounded and dead animals found in burned areas. The collections were accomplished in randomized points in September 2003, during - and immediately - after fire occurrences. Three snakes were observed during wildfires, one Bothrops moojeni, one Philodryas patagoniensis, and one non-identified colubrid. A Micrurus lemniscatus was found alive, presenting deep burns and visual deficiency due to thermal lesions. The corpses of four B. moojeni, one non-identified colubrid, and one Tamnodynastes hypoconia were collected. The four B. moojeni and the non-identified colubrid were carbonized. The T. hypoconia presented signs of respiratory illness. Vestiges indicated that snakes corpses are an important source of food in post-fire periods. Highly intense wildfires which cover large areas directly affect the Ilha Grande National Park snakes, including big species as B. moojeni. Individuals’ death due to fire action can contribute to the snakes species’ declining in the park in a long term basis.


Los incendios son ocurrencias periódicas en el Parque Nacional de Ilha Grande (sur de Brasil), y uno de los principales factores de amenaza a la supervivencia de la herpetofauna en esa unidad de conservación. Los objetivos de este trabajo fueron describir las reacciones de las serpientes al fuego, identificar las especies afectadas por los incendios y correlacionar la naturaleza de las lesiones causada por el fuego. Las actitudes de las serpientes frente al fuego fueron determinadas por la descripción de las reacciones de comportamiento presentadas por los individuos observados durante los incendios. La determinación de la causa muerte y lesiones se efectuó por evaluación física y necroscópica de los animales heridos y muertos encontrados en las áreas quemadas. Las colectas fueron realizadas en puntos aleatorios en septiembre de 2003, durante y inmediatamente después de las ocurrencias. Fueran avistadas frente al fuego, un Bothrops moojeni, un Philodryas patagoniensis y un colúbrido no identificado. Se encontró un Micrurus lemniscatus vivo, presentando quemaduras profundas y deficiencia visual, debido a las lesiones térmicas. Fueron colectados los cadáveres de cuatro Bothrops moojeni, de un colúbrido no identificado, y de un Tamnodynastes hypoconia. Los cuatro B. Moojeni y el colúbrido no identificado estaban carbonizados. El T. hypoconia presentaba signos de enfermedad respiratoria. Vestigios indicaron que los cadáveres son importante fuente de recurso alimenticio en los períodos posfuego. Los incendios de alta intensidad y gran extensión afectan las serpientes directamente en el Parque Nacional de Ilha Grande, incluso animales de talle grande como B. moojeni. La muerte de individuos por acción del fuego puede contribuir para la caída de las especies en el parque, a largo plazo.

ABSTRACT

PURPOSE: The emergence of multiple resistance to antimicrobials in Vibrio cholerae isolated in the state of Ceará, Brazil, alerted researchers in this area to the sensitivity to antimicrobials of strains isolated in Rio Grande do Norte (RN), Brazil. METHODS: One hundred and four strains of V. cholerae of human origin, isolated by Laboratório Central de Saúde Pública Dr. Almino Fernandes, were serologically typified and evaluated for in vitro sensitivity to eight antibiotics belonging to different groups (polymyxine, tetracycline, chloramphenicol, nitrofurantoin, sulphazotrin, pefloxacine, erythromycine, ampicillin). The strains were collected from patients suspected of contracting choleric diarrhea in the year 1999, in Natal/RN/Brazil. RESULTS: From the sample total, 100 were identified as V. cholerae, serogroup O:1, biotype El Tor, with 99 (95.3%) belonging to serovar Ogawa and only 1 (0.9%) to serovar Inaba. The 4 remaining were characterized as non O:1 V. cholerae, with 3 (2.9%) biochemically identified as Heiberg type I and 1 (0.9%) as type II. All the V. cholerae serogroup O:1 strains were sensitive to tetracycline, chloramphenicol, sulphazotrin, pefloxacine, erythromycine and resistant to polymyxine. In relation to nitrofurantoin, only 1 was sensitive. Only 1 was resistant to ampicillin. The non O:1 V. cholerae strains were resistant to polymyxine. CONCLUSIONS: The results showed sensitivity in 100% of the V. cholerae serogroup O:1 strains to tetracycline, an elective drug in the treatment of cholera, and an absence of multiple resistant strains in our environment. An interesting finding was the frequency of serovar Ogawa in 1999, considering the greater incidence of serovar Inaba in other years of cholera outbreaks in RN.
